The Molecular Design and Therapeutics lab offers protein engineering and expression support for local investigators. Our focus is on delivering preclinical quantities of endotoxin-free protein and providing all the expertise needed for the design of novel biologics.

Available Services

Design & Expression of Constructs

  • Design, expression and purification of proteins in mammalian cell lines. Just let us know what you would like to express and we will help design the ideal construct using our customized suite of lentiviral-based expression vectors.
  • Help you clone your constructs into our vectors.
  • Increase the yield of difficult to express proteins using our customized fusion constructs.
    • Express your epitopes as fusions to human, animal or murine proteins for downstream immunizations.

In Vivo Biotinylation

  • In vivo biotinylation of your protein using our Freestyle 293-F cells stably expressing the E. coli biotin ligase enzyme to streamline downstream experiments (e.g. Biacore).
  • Multimerize your proteins using a variety of our customized fusion partners.
  • Pilot-scale (≥ 100mL) and large-scale expression (≤ 6L) and purification of endotoxin-free proteins for animal studies.

Customization Services

Since each protein expression project is unique we will help tailor expression strategies to the unique needs of the investigator. We have all the expertise available to assist investigators in designing expression constructs that maximize success.

  • We routinely express:
    • Full length antibodies (heavy and light chains on separate viruses)
    • Single chain variable fragments
    • Cytokines and other secreted proteins
    • Stable domains of larger proteins
    • Ectodomains of transmembrane proteins

Agilent 1260 HPLC

The Molecular Design and Therapeutics support lab has one Agilent 1260 HPLC equipped with in-line degasser, diode array detector, fluorescence detector and single quadrupole MS. It is capable of fractionating samples for a variety of applications on a semi-preparative scale.

Labconco Lyophilizer

The Molecular Design and Therapeutics support lab has one Labconco -105°C 4.5 liter freeze dry system.

AKTA Pure FPLC

AKTA Pure FPLC

The Molecular Design and Therapeutics core lab has two ÄKTA pure 25 L systems for routine affinity chromatography and one ÄKTA pure 25 M (with multiple wavelength monitoring). Each ÄKTA system is equipped with an external system pump and fraction collector. We also have most chromatography columns used for routine protein separations.

Infors HT Shaking Incubator

Infors HT Shaking Incubator

The Molecular Design and Therapeutics support lab has six Infors HT Multitron shaking incubators capable of handling over 60 liters of suspension culture at one time. We routinely grow Freestyle 293-F cells.
